Pernicious anemia (anemia) is an autoimmune disease that causes a deficiency of vitamin B12. A protein called intrinsic factor, which is usually in the stomach, and is necessary for the absorption of vitamin B12, "missing".

It is not known how vitamin B12 deficiency causing nerve damage. B12 is known to be required in human metabolism as a cofactor of two enzymes to be: one is for the synthesis of methionine, an amino acid that is required, and the other is involved in the degradation of certain amino acids and odd fatty acids.
